炎症性肠病(IBD)是一种难以根治的慢性肠道炎症性疾病,并且有潜在的癌变风险。目前基于小分子药物、抗生素和抗体的临床治疗方法并不完全有效,长期使用甚至可能导致严重的并发症、抗生素耐药性和免疫反应。IBD难以治疗部分原因是治疗缺乏特异性,大多数治疗方式依赖于血液循环很少真正针对病变部位因此可以定位到炎症部位的创新靶向治疗策略非常重要。有炎症结肠的特点之一是富含正电的蛋白质及活性氧(ROS)水平升高,通过带负电荷的药物和药物载体来靶向这些带正电的蛋白质,并清除ROS来减轻炎症反应的治疗策略可能是一种非常有效的治疗方法。


纳米材料在生物成像、癌症早期诊断和药物运载等领域的应用越来越广泛,为医学诊断和治疗提供了有力的物质基础和技术支持,如何安全、有效、方便地使用纳米材料是纳米医学的最大难题之一。纳米二氧化铈是一种具有超氧化物歧化酶和过氧化氢酶样活性且可以清除ROS的多酶模拟物,催化抗氧化活性使纳米二氧化铈能够通过消除ROS而有效地治疗各种疾病,包括炎症性疾病。与CeO2纳米颗粒不同,蒙脱土(MMT)不能通过胃肠道屏障,也不会干扰正常的结肠功能。近期南京大学魏辉教授课题组在“Advanced Functional Materials”发表了题为“An Orally Administered CeO2@ Montmorillonite Nanozyme Targets Inflammation for Inflammatory Bowel Disease Therapy”的文章,结合CeO2纳米颗粒和MMT独特的理化性质,设计了一种可以口服给药系统,用于IBD的治疗获得了显著的效果。基于酶的疗法相比,纳米铈具有稳定性好多酶活性和免疫原性低等优势。CeO2@MMT通过显著下调促炎细胞因子水平和同时上调抗炎细胞因子水平来调节病变组织的局部免疫环境。


本研究使用了欣博盛小鼠IL-1β  IL-10 ELISA试剂盒评价了小鼠结肠组织匀浆促炎和抗炎细胞因子的水平。
