有临床证据表面怀孕期间发生的生理和免疫变化与感染和败血症期间疾病恶化的结果有关,但这些关联是如何加剧炎症的发展还没有被研究过。最近,南方医科大学的研究团队在期刊《Immunity》(IF=43.474)上发表题为 “ Pregnancy-induced changes to the gut microbiota drive macrophage pyroptosis and exacerbate septic inflammation. ” 的文章,该研究使用抗生素治疗和粪便微生物转移,证明脓毒症易感性是由妊娠诱导的小鼠和人类肠道微生物组的变化驱动的。综合多组学和基因工程细菌揭示,怀孕期间Parabacteroides merdae (P. merdae)(副拟杆菌科(拟杆菌科))数量减少导致芒柄花素(Formononetin,FMN)减少和巨噬细胞死亡增加。从机制上证明,FMN通过抑制hnRNPUL2的核积累和随后与NLRP3启动子的结合来抑制巨噬细胞的核下垂。用FMN治疗或敲除鼠hnRNPUL2可预防脓毒性炎症。P. merdae和FMN的肠道丰度与脓毒症患者的进展呈负相关。该篇文章研究成果揭示了微生物免疫轴在妊娠脓毒症宿主中被破坏,突出了FMN-hnRNPUL2-NLRP3轴在为脓毒症的治疗提供有前景的策略和潜力。
